    胎盘特异性蛋白1(PLAC1)是癌症睾丸抗原(CTA)家族的重要成员,具有典型的肿瘤限制性表达特征,在多种恶性肿瘤中异常高表达,而在正常组织中仅见于睾丸和胎盘。研究发现,PLAC1参与肿瘤发生发展过程,成为潜在的诊断标志物和治疗靶点,在头颈部鳞状细胞癌(HNSCC)中亦表现出显著致癌性。基于其高免疫原性和肿瘤特异性,针对PLAC1的抗体偶联药物、多肽疫苗等疗法正在研发中,纳米靶向技术也显示出跨癌种治疗潜力。本文将概述PLAC1的结构特征、表达调控机制及其在肿瘤发生发展中的作用,并探讨其在HNSCC中的生物学功能和治疗前景。

    Abstract:

    Placenta-specific protein 1 (PLAC1) is an important member of the cancer-testis antigens (CTA) family, which exhibits tumor-specific expression patterns. It is abnormally highly expressed in multiple malignant tumors, while it is only found in the testis and placenta in normal tissues. It has been discovered that PLAC1 is involved in the process of tumor occurrence and development and has become a potential diagnostic marker and therapeutic target. PLAC1 also exhibits significant carcinogenicity in head and neck squamous cell carcinoma (HNSCC). Based on its high immunogenicity and tumor specificity, antibody-coupled drugs and peptide vaccines of targeting PLAC1, and other therapies are currently under development. Nano-targeting technology also shows potential for cross-species cancer treatment. This paper summarized the structural characteristics, expression patterns, and regulation mechanisms of PLAC1 expression, as well as its role in tumor occurrence and development. The paper also reviewed its biological function and therapeutic prospects in HNSCC.
